Adjuvant Induced Nitric Oxide Production by Bone Marrow Cells Shows NKT Cell Involvement

How adjuvants enhance immunogenicity is critical for advances to take place in vaccine delivery. In addition, understanding how adjuvants function may help our overall comprehension of inflammation. Injecting incomplete Freund’s adjuvant (IFA) into hind footpads of mice induces a rapid immune response as measured by the ability of bone marrow (BM) cells to produce nitric oxide (NO), in response to lipopolysaccharide (LPS) and interferon-gamma (IFNγ). The increase in NO production occurs within 48 hours of injection and is gone by day 6. Also there are higher levels of NO produced when BM cells from IFA injected mice are cultured with anti CD3 antibody and LPS. These increases are not present in NKT cell deficient mice that lack MHC class 1 and 1b molecules. The data shows that IFA induces a rapid response in the innate immune system as measured by NO production and this response is absent in NKT cell deficient mice. These findings suggest that activation of NKT cells cause enhanced NO production, which can be associated with pathogenic affects such as inflammatory osteoporosis.
