Expression of Kruppel-like factors 6 & 7 in Central Visual Structures of Adult Zebrafish Following Optic Nerve Crush

Kruppel-like factors (KLFs) are zinc finger proteins which bind to promoter or enhancer regions of genes in order to regulate their transcription. They are involved in many functions including the regeneration of retinal axons following optic nerve damage. However, studies involving optic nerve regeneration in zebrafish have only surveyed Klfs expression and function in retinal ganglion cells. In this study I used in situ hybridization to determine if expression of Klf6 and Klf7 were upregulated in the optic tract and/or optic tectum (the major target for the retinal axons) following damage to the optic nerve in adult zebrafish. Klf6 and Klf7 were both upregulated 1-day post-injury (dpi) in the optic tract, while increased Klf6 expression was also detected in the retinorecipient regions of the optic tectum. Expression levels of these two Klfs (based on staining intensity) became further increased in both the optic tract and the optic tectum of fish 2-7 dpi. In the optic tract and optic tectum of fish 14 dpi, expression of Klf6 and Klf7 continued to be elevated compared to their respective control levels, but reduced compared to that of 2-7 dpi. By 21 dpi, expression of both Klfs in the optic tectum became similar to their respective controls, but remained increased in the optic tract.
