LOCALIZED ANTIBIOTIC DELIVERY VIA VALINE BASED POLY(ESTER UREA)

Despite advances in aseptic surgical techniques and prophylactic antibiotics, the incidents of device associated infections are rising dramatically along with the increasing clinical demand for implanted devices. In this study we have attempted to tackle one type of device associated infection, the cardiac implantable electronic device (CIED) infection. The objective of this study was to design an antibiotic loaded pouch for CIEDs from novel valine based poly(ester urea) polymers for localized antibiotic delivery of antibiotic. Implanting CIED covered in this pouch would help prevent infections even at antibiotic dosage lower than currently employed. Additionally, it is expected to reduce side effects of systemic antibiotic administration and put minimum pressure on hepatic and renal functioning of patients. In this study, films with different antibiotic loading concentration and thickness were fabricated and analyzed for their antibiotic release profiles in vitro. Furthermore, antimicrobial susceptibility tests were also performed to confirm that antibiotic released from these groups retained its antimicrobial activity.
