EFFECT OF AGING ON ESTRUS CYCLING AND MEMORY IN SENESCENCEACCELERATED MOUSE (SAMP8)

Aging is associated with a decline in memory. In females this decline may be associated with reduced levels of estrogen. To date, most studies investigating age-related changes in memory have used male animals. The present study examines the effects of age on estrous cycles and spatial memory in female senescence accelerated mice (SAMP8). Four ages of SAMP8 were examined: 2-months, 5-months, 7-8 months, and 12-months. Spatial memory was assessed by examining performance on a water maze task (5 days, 2 sessions/day, 4 trials/session). Stage of the estrous cycle was determined by performing a vaginal lavage and examining the cells daily to determine the regularity of cycling. Vaginal smears were taken over a period of 12 days, and viewed microscopically for classification. The water maze task showed that performance was similar for the 2- and 5-month mice and for 7- and 12-month mice. Furthermore, memory of the two older ages was impaired compared to the two younger ages. With respect to the effect of age on estrous cycle in SAMP8, duration of cycling was 3 days at 2-months, 4 days at 5-months, 5 days at 7-months and 6 days at 12 months and older, showing a gradual lengthening of the estrus cycle with increasing age. The present study does not support a simple relation between lengthing of the estrous cycles and memory impairment in aged SAMP8 female mice.
