The Effect of PCB 47 and PCB 77 on the Female Reproductive System: Gestational Versus Lactational Effects in Rats

Polychlorinated biphenyls (PCBs) are a group of chemicals that has been widely known for their endocrine disrupting effects. Because of their commercial use in industries all over the world, they have found their way into the environment and remain a concern for the health of public. One of the major disrupting effects of PCBs is the disturbance of the reproduction system of mammals. To investigate the disruptive effect of a particular mixture, PCB 47/77, on young rats and also to elucidate the different disruptive effects caused by the differences of exposure stage in these developing animals, in the present study pregnant rats were fed diet containing the PCB47/77 mixture and their pups were cross-fostered to mothers not exposed to PCB and vice versa. Our result have shown that the PCB 47/77 treatment can induce disrupting effect on the reproductive system of their female pups, causing the prematuration of the ovaries and uterus as early as 17 days of age. The group of cross-foster pups exposed to PCB lactationally appears to suffer more greatly from various disruptive effects than those exposed gestationally, suggesting that the neonatal developmental stage could possibly be even more vulnerable than the gestational period of development. Since this trend is consistent with a significantly lower concentration of serum thyroid hormone than in the other cross-fostered group, it is likely that hypothyroidism plays a major role in the PCB disrupting effects.
