Collecting Low-level DNA Sample of Foreign Contributor From Skin of Known Sources

Baughman, Joan Nicole

Abstract Details

2019, Master of Science (MS), Bowling Green State University, Forensic Science.
This study was designed to test the hypothesis that full touch DNA profile of a foreign contributor can be extracted from the skin of a participant. Through trial and error, multiple procedures were performed to attempt to obtain the full profiles. The most efficient method was a combination of the GlobalFiler PCR Amplification Kit, the PCR protocol for the thermal cycler and a modified version of the lysis procedure used in the GlobalFiler PCR Express Amplification Kit. The results throughout were inconsistent, inconclusive and the procedure was unable to be completed properly due to manufacturer level contamination of the GlobalFiler PCR Amplification Kit. New kits need to be purchased and improved modified procedure needs to be replicated. After this occurs, the results should be analyzed and compared as was the previous intention.
