Abstract
The present studies have used several approaches to understanding an essential point of eukaryotic gene regulation: what controls the exchange of macromolecules between the nucleus and cytoplasm? Three approaches are presented. In the first, a panel of yeast mutants are identified that accumulate poly-adenylated mRNA within the nucleus at restrictive temperatures, implying a lesion in mRNA export. The genetic basis of an individual mutant is due to a lesion in the MTR13 gene, which codes for an ubiquitous nuclear protein with an RNA binding domain and an extensive arginine-glycine repeat domain. The arginine-glycine repeat domain is similar to domains found in a number of nuclear proteins involved in RNA metabolism. A mutation in the gene was found to block both RNA export and protein import into the nucleus. The mtr13 mutant is the first yeast mutant for which a bidirectional block in nucleocytoplasmic transport has been shown. In the second approach, expression of a gene containing extensive predicted secondary structure has detrimental effects on the growth of otherwise wild-type yeast, suggesting a possible way of screening for suppressor mutations that might be involved in the RNA export pathway. In the third approach, a biochemical method is presented for obtaining nuclear membranes, in order to more carefully characterize the proteins of the inner nuclear envelope
