Abstract
Hemocyanins are extracellular copper containing oxygen transport proteins found some arthropod and molluscs. Limulus polyphemus hemocyanin is a 48 subunit complex, each subunit can reversibly bind one oxygen molecule. (Volbeda, A. and Hol, W. G. L., Crystal Structure of Hexameric Haemocyanin from Panulirus interruptus Refined at 3.2 A, J. Mol. Biol. 209, 249-279, 1989) shows that the two proteins have similar three-dimensional structures. However there is a significant difference between the position of the amino terminal domains of the two arthropod hemocyanin types. The three-dimensional structure of purified Limulus polyphemus subunit II hemocyanin in the oxygenated form has been determined by single crystal X-ray diffraction methods. The structure was determined to 1.9 A resolution to a crystallographic R-value of 18.6%. An oxygen molecule is well-ordered in the crystal and appears in an μ-η2:η2 configuration with the two copper atoms being 3.6 ± 0.2 A apart. No endogenous protein bridging ligand is in the immediate vicinity of the two copper atoms. A path by which ligands may reach the active site from the surface of the subunit to the buried dicopper oxygen binding site is indicated.
