A Human Genome Epidemiology Systematic Review of Endothelin Receptor-A

The aims of this project were two-fold. The existing guidelines for HGE systematic review were examined and novel approaches to this process were proposed in Phase I of this project. Suggestions included organizing reviews by gene, analysis of data by linkage disequilibrium block, and the use of an audit tool to guide data mining. In Phase II, these advanced guidelines were implemented in a review of endothelin receptor-A (EDNRA). A five database literature draw-down yielded 267 unique abstracts for review. From these, 79 full-text studies were obtained; 24 studies met eligibility criteria. Analysis of the data was performed using the Venice Criteria (Ioannidis et al., 2008) and amalgamation by linkage disequilibrium block. While the Venice Criteria proved too stringent for the emergent EDNRA data set, analysis by linkage disequilibrium block yielded several trends including stratification of association by sex within block 1, synergistic associations with variants of endothelin-1 in block 3, and fairly strong associations with hypertension/blood pressure outcomes in block 4. This study provides advanced guidelines for HGE systematic review and evidence of their usefulness in practice.
