Surface Drying and Rehydration Does Not Affect the High Cycle Fatigue Behavior of Human Bone Tissue

Raman spectroscopy allows for non-destructive analysis of bone tissue composition and quality. The use of Raman spectroscopy to predict the resistance of bone tissue to cyclic loading has not been prospectively established. A 2-hour surface drying protocol was proposed for Raman assessment of bound water compartment. The objective was to determine if a surface drying and rehydration protocol affects the cyclic fatigue life of human cortical bone. Twenty-four specimens were distributed into Hydrated group and Surface Dry / Rehydrated group and were tested under high-cycle fatigue conditions at 25, 35, 45, and 50 MPa in a rotating bending test frame. Raman analysis were conducted under both conditions. No difference in high cycle fatigue behavior was found between the Surface Dry / Rehydrated group and the Hydrated group in statistical analysis and fracture surface analysis. This study also revealed differences in fatigue life for different anatomical quadrants.
