MULTIOMICS EVALUATION OF THE EFFECT OF EZRIN INHIBITOR IN DIFFUSE LARGE B CELL LYMPHOMA REVEALS DYSREGULATION OF BCR SIGNALING, RHO GTPASE SIGNALING AND APOPTOSIS

Activated B-cell like diffuse large B-cell lymphoma (ABC-DLBCL) is one of two molecular subtypes of DLBCL. ABC-DLBCL exhibits worse outcome following standard therapy. The pathogenesis of ABC-DLBCL is due to constitutive activation of NF-κB signaling pathway which lead to activation of numerous of processes like cell proliferation and survival. Recently, findings indicate ezrin inhibitors (Ez-Is), NSC305787 and NSC668394, were found to disrupt BCR signaling and decrease pro-survival signaling through induction of apoptosis. To gain more insights into the mechanism of action of Ez-Is in ABC-DLBCL, we performed a multiomics analysis of RNA microarray, protein and phosphoprotein mass spectrometry (MS) from ABC-DLBCL cell lines treated with Ez-I. Our results revealed Ez-I affect many genes, protein and phosphoproteins. The pathway enrichment of them showed pathways mainly related to BCR signaling, Rho GTPase signaling and apoptosis are affected. Our findings were similar to published data and increased our understanding of Ez-I in ABC-DLBCL.
