Application of Affinity Polymer Coating to ePTFE Arteriovenous Grafts to Improve both Passive and Active Infection Prevention for Hemodialysis

During hemodialysis treatment, infection in arteriovenous grafts results in severe consequences. Patients often have to have the complete graft removed, causing additional surgeries and other long-term complications. This project aimed to improve infection prevention in arteriovenous grafts by applying a cyclodextrin polymer coating to expanded polytetrafluoroethylene (ePTFE) vascular graft material. Different coating solutions were applied to grafts to identify optimal conditions for coating. Methylene blue dye and SEM imagery was used to assess success of cyclodextrin attachment. Coated and uncoated grafts were compared in three areas: drug release kinetics, bacteria killing via a zone of inhibition assay, and bacterial adhesion prevention. In all three areas, coated grafts outperformed uncoated material. Both cyclodextrin’s non-specific affinity binding and hydrophilic nature allows for a combination of active and passive infection resistance. With this new coating, infection may be better prevented, decreasing the amount of complications associated with AV synthetic grafts.
