INTERACTIONS AND LOCALIZATION OF PROTEIN PHOSPHATASES, YWHA PROTEINS AND CELL CYCLE CONTROL PROTEINS IN MEIOSIS

Oocytes, the female gametes, are produced during oogenesis and held at meiotic prophase I arrest in the ovary until reproductive maturity. In mammals, the oocyte must undergo maturation to differentiate into the fertilizable egg. It is known that a key protein involved in resumption of meiosis is Cell Division Cycle 25B (CDC25B), which dephosphorylates and activates Cyclin-Dependent Kinase I (CDKI). Active CDKI initiates germinal vesicle breakdown (GVBD), the first signal of resumption of meiosis. The tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation proteins (YWHA) are known to be responsible for maintaining arrest through binding CDC25B, rendering it inactive. Here, co-localization analysis and FRET were used to show that YWHAH and YWHAE are able to form heterodimers in the mammalian oocyte. Members of the phosphoprotein phosphatase family of serine/threonine phosphatases (PPP) are also implicated to be involved in regulation of CDC25B. Through various pull down assays, it is observed that PP1¿1 binds CDC25B in the ovary. When PP1¿ is absent, PP1a binds CDC25B, implicating these isoforms may be functionally interchangeable. This works lays important foundations for the future study of the intricate pathway involved in mammalian female meiosis.
