FATTY ACID-LIKE Pt(IV) PRODRUGS HARNESSING CD36 TO TARGET CHEMORESISTANT OVARIAN CANCER CELLS

The long-standing challenge of treating ovarian cancer is drug resistance to the conventional platinum drugs. CD36 is a member of the class B scavenger receptor family of cell surface proteins and responsible for facilitating uptake of free fatty acids for lipid metabolism. Recent studies indicate that CD36 is upregulated in ovarian cancer via interacting with adipocytes. The extracellular domain of CD36 has a deep hydrophobic pocket that can reversibly bind and release free fatty acids, which facilitates the active transport of these amphiphilic molecules across the cellular membrane. Even though the role of CD36 in mediating cellular uptake of small molecules and biomacromolecules is well documented, its capability in shuttling therapeutics contents remains unknown. This thesis presented a study of fatty acid-like Pt(IV) prodrug harnessing CD36 to target drug-resistant ovarian cancer cells. These are based on the studies on the CD36- dependent cellular uptake, cytotoxicity and CD36 expression in different cell lines. The results show the fatty acid-like Pt(IV) prodrugs efficient target chemoresistant ovarian cancer cells via CD36.
