Previous work has shown that male oxytocin receptor (Oxtr) knockout (KO) mice show increased aggression in adulthood. Previous work has also indicated the serotonin 1B receptor to be important for the inhibitory modulation of aggression across rodent species. We hypothesized that the increase in aggression seen in Oxtr KO mice is at least partially mediated by the serotonin system. CP-94,253 (10mg/kg), a selective serotonin 1B receptor agonist, was used in a resident-intruder aggression test to test this hypothesis. CP-94,253 was found to reduce aggression. Due to apparent increases in aggression from the injection of the drug across groups, the data across all 3 days of testing is uninterpretable. However, examination of day 1 data reveals a specific reduction of aggressive behavior in the CP-94,253 treated Oxtr KO animals. This study provides preliminary evidence showing that the increase in aggression seen in adult Oxtr KO mice may be due, at least in part, to altered serotonergic tone.
