Neuroanatomical and Cellular Localization of Luteinizing Hormone in the Mouse Brain

Previous work in our laboratory demonstrates the importance of luteinizing hormone (LH) signaling in learning and memory and Alzheimer's disease (AD). Though we have also previously demonstrated that LH is present in the brain, the neuroanatomical and cellular localization of LH has not been determined. To address this, we carried out immunofluorescence to determine in which brain regions and cellular types LH are located. Immunoreactivity of LH is observed in limbic system areas associated with emotion as well as learning and memory and several midbrain motor nuclei. Importantly, high expression of LH is visualized in the retrosplenial cingulate cortex and areas of critical importance in AD. We have previously also shown that LH co-localizes with NeuN a neuronal marker, however, whether LH co-localizes with excitatory or inhibitory neurons is not known and important in understanding the role of this hormone in regulating neuronal circuits. To address this, we carried out immunofluorescence to determine in which cells LH co-localized with. Expression of LH co-localized with GAD67 an inhibitory neuron marker. Given that several subtypes of inhibitory neurons also exist, to address which type of inhibitory neuron LH co-localized with we also determined co-localization of LH with the three most common types of inhibitory neurons: parvalbumin, somatostatin, and vasointestinal peptide expressing neurons using mice carrying a fluorescent reporter for these cells or double-labeling immunofluorescence. Our data demonstrate that LH did not co-localize with any of these subtypes of inhibitory neurons. These data therefore suggest that 1) LH co-localizes with other subtypes that we have no analyzed or 2) we have identified a novel LH expressing inhibitory neuron. Future studies will evaluate these aspects in more depth.
