Synthesis, Purification, and Structural and Dynamic Studies of the Amino-Proximate Transmembrane Domain of CREP-1, a Diverged Microsomal Delta-12-Desaturase

The plant membrane-bound acetylenase/desaturase CREP-1 was studied using solid-state NMR spectroscopy and CD spectroscopy. Desaturases dehydrogenate fatty acid derivatives. The functionally diverged desaturase, CREP-1 and its homologs carry out diverse oxidative transformations on unsaturated fatty acids. The structural characteristics of the desaturases are, nevertheless, not well described. CREP-1 is thought to serve as a good model system for desaturases, and structural analysis of this enzyme will facilitate a better understanding of desaturases. The region of CREP-1 investigated in this initial study was the amino-proximate transmembrane domain (TM-A). TM-A has been synthesized with a peptide synthesizer using Fmoc chemistry and purified by reverse-phase HPLC. It was shown to be 50-70% alpha-helical by CD spectroscopy under a variety of conditions. Solid-state 15N and 2H NMR data showed this peptide to be transmembrane in a phospholipid bilayer. Solid-state 2H NMR data defined the residues that were buried in the bilayer and solvent-exposed by examining side-chain dynamics. Other regions of CREP-1 are being studied using analogous methods. The compilation of the spectroscopic data from TM-A along with other regions of CREP-1 being studied should provide a good overall structural picture of CREP-1 and serve as a model for future work on desaturases.
