Methylammonium Formate as a Mobile Phase Modifier for Reversed Phase Liquid Chromatography

This study focuses on the synthesis and the characterization of the room temperature ionic liquid methylammonium formate (MAF) and its use as an organic solvent replacement for the mobile phase in liquid chromatography. Spectroscopic characterization as well as measurements of physical properties are determined and compared to previously studied alkylammonium formate ionic liquids. Our chromatographic evidence has shown that MAF is a good silanol suppressor resulting in better peak shape and separation efficiency than methanol as a mobile phase modifier when using C18-based silica columns. The preliminary studies using MAF as a mobile phase modifier or additive for pharmaceutical applications using LC-MS methods have shown promising results. The fluorescence studies using cytochrome C as the test compound in five different organic solvents has identified MAF as the least denaturing compared with other solvents including dimethyl sulfoxide, N-methylformamide, methanol, and acetonitrile.
