In this thesis thermodynamic properties of NADH (dihydronicotinamide adenine dinucleotide) conformational states at 1 atm pressure were characterized in a pH buffer MOPS (pH = 7.5) with varying concentrations of denaturant (methanol) concentrations. Linear model and spectral decomposition model were developed to calculate fraction-unfolded values of NADH in different methanol concentrations. An optical sectioning system using a structured illumination setup based on previous literature was built for use with a wide-field microscope. The sectioning ability of the system was shown in slide and capillary mounted paper fibers. The illumination setup was also used to a acquire sectioned images by varying the z-axis coordinates and building a 3D image of the slide mounted paper fibers, and used to show a comparison between the conventional and sectioned 3D image.
