The Effects of Cold Acclimation on the Thermogenic Capacity of Skeletal Muscle in Mice Deficient in Brown Adipose Tissue

UCP-dta mice have a reduced capacity for non-shivering thermogenesis due the partial ablation of brown adipose tissue, but despite this deficit, they are not cold intolerant. Therefore, we hypothesized that UCP-dta mice would rely on shivering thermogenesis in skeletal muscle during a cold challenge, therefore resulting in a higher muscle aerobic capacity. The VO2summit of UCP-dta mice acclimated to standard facility temperatures was lower than wild type controls. Cold acclimation caused cardiac hypertrophy and increased brown adipose tissue mass, VO2summit, and citrate synthase activity in both wild type and UCP-dta mice. However, brown adipose tissue mass and VO2summit were still lower in cold acclimated UCP-dta mice relative to the cold acclimated wild type mice. Because cold acclimation increases skeletal muscle oxidative capacity nearly equivalently in wild type and UCP-dta mice, and VO2summit is still compromised in UCP-dta mice, shivering thermogenesis does not compensate for brown adipose tissue deficiency.
