DEVELOPING HOMOGENEOUS BRAIN-MIMICKING CRYOGELS FOR MAGNETIC RESONANCE ELASTOGRAPHY

Magnetic resonance elastography (MRE) is a noninvasive technique that is used to quantify the shear stiffness of biological tissues for diagnostic assessments (e.g., tumors or traumatic brain injury). Currently MRE is used at Mayo clinic and has been approved by the U.S. Food and Drug Administration (FDA) for diagnostic assessment of liver. The feasibility of MRE for other organs such as brain has been proven; however, it requires many clinical trials involving a high number of patients to obtain adequate statistical significance. Therefore, a tissue-mimicking phantom that exhibits the same mechanical properties as those of brain tissue is necessary for validation purposes. Polyvinyl alcohol (PVA) cryogel prepared by the freeze-thaw technique is a potential candidate for mimicking the mechanical properties of soft tissues and has been extensively used as a phantom material. Direct mechanical characterization of PVA cryogel phantoms will allow verifying the accuracy of the shear modulus measured using MRE technique. ABAQUS software has been employed in this study for predicting the heat transfer within the phantoms, in an effort to develop homogeneous cryogels. Chapter 1 of this thesis presents the principles of MRE technique. In addition, this chapter provides the details of the experimental procedure for producing homogeneous PVA cryogels by freezing/thawing method. Chapter 2 and 3 give the submitted manuscript based on this study and a published joint work, respectively. The last section includes the conclusions and suggestions for future work.
