Spectrophotometric Determination of Nitrite by Derivatization with Captopril

Nitrite level determination is important for monitoring drinking water quality. For this study nitrite is detected in water by its reaction with captopril, a molecule originally developed to treat hypertension. Sodium nitrite rapidly nitrosylates captopril under acidic conditions, which produces a derivative with concentration-dependent absorption bands at ~ 333 nm and ~ 546 nm. These bands provide overlapping linear dynamic ranges that enable nitrite determination across ~ three orders of magnitude under a single set of experimental conditions. This reaction is complete within 40 minutes at room temperature at an optimum pH of 3. Two interference studies are conducted. Interference effects by various environmentally relevant cationic species and anionic species are found to minimally affect product formation. The amino acids alanine and cysteine were found not to interfere. However, an interference is found in the presence of lead and ionic strength can be a factor due to the decrease in absorbance intensity. These results highlight the potential utility of the captopril-based spectrophotometric assay for sensitive, selective nitrite determination.
