Fluorescent derivatization of glycine using o-phthalaldehyde and captopril for the indirect determination of nitrite

In this research, we developed an indirect fluorescence method for determination of nitrite based on a two reaction sequence. The nitrosocaptopril produced by the first reaction between nitrite and excess captopril is not fluorescent. The fluorescent product produced by the residual captopril with glycine and o-phthalaldehyde (OPA) is then measured and related to the nitrite concentration. During the development of the method, we optimized comprehensively the reaction condition parameters, including the thiol regent for the two reactions, reaction temperature and pH value for the first reaction, and the choice of amino acid, pH value, the ratio of reactants, buffer concentration, reaction temperature, and time for the second fluorescence reaction. Under the optimized conditions, we investigated the effect of eight cations and ten anions on the fluorescence value. The results indicated there was good selectivity except for iodate and periodate, which could be removed by addition of a reducing agent. For the analytical figures of merit, the limit of detection and quantitation of nitrite were 0.8 nM and 2.5 nM, respectively. The fluorescence intensity was found to linearly decrease with increased nitrite from 0 nM to 140 nM. The method has shown to have good inter-day and intra-day precision.
