Structural Studies of KCNE1 in Lipid Bilayers with Magnetic Resonance Spectroscopy and Characterization of Membrane Mimetic Lipodisq Nanoparticles

In general, structural studies of membrane proteins are more difficult than soluble proteins mainly due to their intrinsic hydrophobic properties. The lack of suitable biophysical techniques and proper membrane mimetics has greatly hindered the research of membrane proteins. In this dissertation, efforts have been made on the development of new biophysical techniques and membrane mimetics using the model membrane protein KCNE1. Chapter 2 focuses on utilizing 31P solid-state NMR spectroscopy to examine the changes in dynamic properties of synthetic lipid bilayers upon incorporation of the membrane protein KCNE1. Chapters 3 and 4 aim at the characterization of a newly-developed membrane mimetic termed lipodisq nanoparticles by dynamic light scattering (DLS), solid-state NMR spectroscopy, and transmission electron microscopy (TEM) techniques. This alternative membrane mimetic broadens the choices of the synthetic lipid systems for structural studies of membrane proteins. Chapters 5 and 6 successfully demonstrate the application of electron spin echo envelope modulation (ESEEM) spectroscopy coupled with site-directed spin-labeling (SDSL) for the local secondary structure determination of the membrane protein KCNE1 in both micelle and lipid bilayer environments. This advanced pulsed EPR technique provides a fast and straightforward method which can be adapted to the study of membrane proteins with unknown structures in a native-like environment.
