DEVELOPMENT AND APPLICATIONS OF HPLC-MS/MS BASED METABOLOMICS

High-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) is a powerful and mainstream analytical platform widely used in both targeted and untargeted metabolomics analysis, thanks to its high sensitivity, selectivity, and reproducibility. Therefore, it was chosen as the major technique of our research and was used throughout my thesis projects. In this thesis work, the development of metabolomics techniques from targeted analysis to hybrid metabolomics approach, has been introduced. This thesis starts with an introduction focused on the general workflow of hybrid metabolomics and the analytical considerations of the optimized hybrid approach. This review is then followed by chapter 2 and 3, two published studies that demonstrated the utility of targeted metabolomics methods to different biomedical applications. Specifically, chapter 2 compared the impact of ultrafine particles from petrodiesel and biodiesel combustion to gut bacterial metabolism; chapter 3 applied targeted metabolomics to differentiate metabolic syndrome from obese patients who did not qualify as be metabolic syndrome via their metabolic profiles. In chapter 4, a hybrid metabolomics workflow was developed to integrate time staggered and mass staggered ion lists with globally optimized targeted-mass spectrometry (ts/ms-GOT-MS); a broader metabolome coverage with an improved analytical performance was achieved.
