Investigating the Dynamic Properties and Structural Topology of Membrane Protein KCNE3 with EPR Spectroscopy

KCNE3 (MiRP2) is a membrane protein that modulates the voltage-gated potassium ion channel alpha subunit KCNQ1. Interaction between these two proteins form a leaky channel that maintains ion homeostasis in the cells. This protein is biologically important since it involves in several fluid and ion homeostasis related diseases. This study mainly focused on probing structural dynamic and topology of KCNE3 in lipid bilayer. Successfully expressed, purified, spin labeled KCNE3 was incorporated into POPC/POPG liposomes. CW-EPR spectral line shape analysis and power saturation techniques were used to study KCNE3 dynamic and topology in lipid bilayer respectively. Inverse central line widths and rotational correlation times of CW-EPR spectra were used to analyze the protein dynamic quantitatively. Topology study of KCNE3 was conducted using power saturation depth parameter data.
