Design and Synthesis of Stable Glucose Uptake Inhibitors

There are many mechanisms that can be targeted when designing anticancer compounds. Due to the Warburg effect, the different steps of glycolysis have been potential targets in recent studies. In order for glycolysis to occur, glucose must first enter the cell via glucose transporters (GLUTs). There have been compounds that effectively inhibit GLUTs leading to reduce cell viability. Unfortunately, some of the most effective compounds discovered are not metabolically stable or exhibit low selectivity. As a result, a series of stable derivatives needed to be synthesized. The general structure of the previously discovered compounds and the new derivatives is similar in that there is one central aromatic ring with one or two pendent phenolic rings attached. The majority of the derivatization occurred at the linkage sites between the central ring and pendent phenolic rings and the placement of functional groups about the pendent phenolic rings. Of the new, more stable derivative prepared many of those compounds exhibit glucose uptake inhibition and/or cell growth inhibition values similar to the less stable compounds previously discovered. The derivatives that performed the best in vitro were those with large aliphatic substitution on the pendent rings.
