The Role of E2F Activators in mouse Development and Tumorigenesis

Mutation of Retinoblastoma (RB) or components that negatively impact this pathway have been identified in different types of tumors. E2F family members, transcription factors regulated by Rb that traditionally have been linked to cell cycle control, are also frequently misregulated in cancers. Interestingly, some tumors have both Rb mutation and overexpression of selected E2F family members, raising the possibility that E2Fs may be performing functions beyond those controlled by Rb. Consistent with this notion, expression profiling has revealed a wide spectrum of E2F target genes that include not only cell cycle regulatory components, but also components that play key roles in the control of differentiation, metabolism, survival, and DNA damage. Our studies using knockout and transgenic techniques suggest that evolutionarily conserved functions of E2F involved in development may be unrelated to cell cycle control and that classical cell cycle functions of E2F identified in cancer may reflect more recent inventions during evolution. Whether cell cycle dependent and independent functions are relevant to the cancer phenotype remains to be experimentally determined.
