Abstract
Tuberculosis, an infectious diseases caused by Mycobacterium tuberculosis, kills approximately two million people annually. Surfactant proteins A and D which combat infections including tuberculosis are hydrophilic proteins produced by type II alveolar epithelial cells. Genetic studies have demonstrated considerable level of polymorphisms in surfactant proteins A and D and they have been found to associated with risk for tuberculosis. In addition to the polymorphisms of SP-A and SP-D, the levels of SP-A and SP-D also vary among individuals. As SP-A and SP-D act in opposite roles in M.tuberculosis infection, we address the question how the ratio of SP-A to SP-D affects the susceptibility to infection with M.tuberculosis. In this paper, a mathematical model describing human tuberculosis at the site of infection in the lung is present in terms of a system of ordinary differential equations. The model was simulated with MATLAB using ode15s and Runge-Kutta 4th order method over the period of 50 days. The model shows that as the ratio of SP-A to SP-D increases the density of healthy macrophages decreases at day 50. On the other hand, the density of infected macrophages, at day 50, first increases with the ratio but when the ratio exceeds certain level the density of the infected macrophages begins to decrease while the total number of macrophages remains essentially the same. The model also exhibits increasing bacterial load as the ratio increases. We conclude that as the amount of SP-A increases relative to that of SP-D, the host becomes more susceptible. This suggests that a drug that decrease the ratio of SP-A to SP-D could be used for the protection against infection with M.tuberculosis.
