Abstract
Pulses of gonadotropin releasing hormone (GnRH) from the hypothalamus stimulate the release of luteinizing hormone (LH) and follicle stimulating hormone (FSH) from the pituitary gland and they control the menstrual cycle and ovulation. This system is affected by feedback from the ovarian hormones, estrogen (E2), progesterone (P4), and inhibin (Ih), which have been shown to have direct effects on the hypothalamus and pituitary in rat, human, and ovine experiments. The hormone E2, P4, and Ih have been shown to change the number of GnRH receptors on pituitary cells, and biphasic responses to E2 on pituitary cells were also reported. We have created a mathematical model that incorporates a previous pituitary-ovarian model and a previously detailed model of the synthesis of LH and FSH in the pituitary so that we can study the dynamics of the whole hypothalamic-pituitary-ovarian axis. The model allows us to compute the level of LH release in response to GnRH pulses when pulse amplitude, duration, receptor numbers, and effective cell numbers are varied, and it enables us to examine the consequences of positive and negative feedback from the ovarian hormones. Our hypothesis is that GnRH receptor number on pituitary cells and the negative feedback on the pituitary cells from E2 are key factors that control the LH surge leading to ovulation.
