Structural Modification of the Flavanone Naringenin – Potential Impacts in Dietary Polyphenol Research

In recent years, there is growing evidence that plant-derived polyphenols influence pathology and symptomology of many high-risk diseases and conditions, including obesity, type 2 diabetes mellitus, cancer and aging. The efficacy of polyphenols, specifically flavonoids, has been extensively studied in in vitro models and are currently being studied in animal models of diseases. However, the translation of biological effect from cell to in vivo models are often limited, due to multiple environmental and host factors. One of most influential factors of polyphenol potency is bioavailability. Studies reveal that many polyphenols are poorly absorbed and retained in humans. Therefore, my thesis research focused on modifying the structure of flavonoids to improve the bioavailability and subsequent activity of the compounds. As previous research has found that prenylation of phytochemicals improves retention capacity, my research focused on similar modification of the flavanone, naringenin. Here, I present a feasible process to synthesize 8-prenyl naringenin from commercially available naringenin by Claisen rearrangement. 8-prenyl naringenin has been observed to be more readily available that naringenin while having compound-specific activity in animal models of diseases.
