Using Tunable Lens to Extend the Range of the Single-Particle Tracking Microscop

Bead tracking with microscope is widely applied in biology, physics, material science especially when people want to know more about the micro world. The facility in my lab can reach high accuracy and resolution of tracking, however, the effective range of measurement in z-position is restricted due to the limitation of microscope itself. Hence, in this research, the design of a new method to increase the measurement range of the microscope is put forward. One piece of focus tunable lens is added to the objective lens to increase the motion range of the lens. When the input current into tunable lens changes, its optical power changes at the same time. In addition, in order to control the focus change of the tunable lens, a controller is designed specifically. Basically, the results derived from the microscope not only show the distance between the z-position of the bead to be measured and the z-position of the objective lens, but also provide input for the tunable lens. Finally, putting the results from microscope and those from the sensor of the tunable lens together, the actual z-position of the bead is obtained with higher range than the original facility. The process of designing and the effective static and dynamic range will be described in detail in my thesis paper.
