Abstract
In this work a prototype on-chip micro-heater for thermocycling is demonstrated, which is the main component for a disposable lab-on-a-chip device than integrates a module for performing polymerase chain reaction (PCR). Several resistive micro-heaters were fabricated from gold/chrome metal films on borosilicate glass wafers and gold metal films on cyclic olefin copolymer (COC) and polycarbonate (PC) plastic wafers. Multiple resistive heaters of different geometries were designed, fabricated, and characterized to explore the feasibility of the micro-heater for a fully integrated disposable lab-on-a-chip with PCR module. Each device was characterized using custom-designed PC-based Labview virtual instruments with a data acquisition and control card. A feedback control program was implemented in Labview to characterize the dynamic response of the prototype thermocycler. The integrated on-chip micro-heater shows that the temperature transitions are short by comparison with the conventional PCR temperature routines, which would allow the fully integrated PCR module to have fast thermocycling. Optimization of the heating capacities of the on-chip micro-heater should be further investigated to make the prototype on-chip micro-heater useful, although a prototype on-chip micro-heater has been designed, fabricated, and successfully characterized to demonstrate feasibility for a fully integrated disposable PCR module.
