DOSE-RESPONSE OF LOW DOSE CO-EXPOSURES TO ARSENIC AND BENZO[a]PYRENE IN MICE

Workers are simultaneously exposed to benzo[a]pyrene (BaP) and arsenic in many industrial settings. Independent epidemiologic studies have reported varying effects—from additive to multiplicative—among such populations. In vitro experiments have shown evidence of synergism between the two exposures, and an in vivo experiment has recently supported this finding. Our goal was to better characterize this relationship using a mouse model with DNA adducts as a measure of effect. Eighty-four female mice were divided into nine treatment groups: +100 nM BaP/- As; +100 nM BaP/+ 2.0 mg As; +100 nM BaP/+ 4.0 mg As; +100 nM BaP/+ 8.0 mg As; +200 nM BaP/- As; +200 nM BaP/+ 2.0 mg As; +200 nM BaP/+ 4.0 mg As; +200 nM BaP/+ 8.0 mg As; and control. Arsenic was provided ad libitum for four weeks in order to reach steady state levels. BaP was topically applied to shaven dorsal skin the four consecutive days prior to sacrifice. Lung and skin tissues were analyzed for DNA adducts using 32P-postlabeling. Inductively Coupled Plasma Mass Spectrometry was used to determine arsenic concentrations in these same tissues. Our results were inconclusive. Relative adduct levels (RAL) in the 100 nM BaP treatment groups were extremely variable. Data for the 200 nM BaP treatment groups were in better agreement with earlier studies, but none of these relationships were found to be statistically significant. Working with such low doses may have been problematic. Actual arsenic concentrations among the 2.0 mg and 4.0 mg treatment groups were very similar, and a greater difference in dose should be used in the future to observe truly heterogeneous treatment groups. Additionally, the 100 nM BaP treatments may have been too low to see a uniform effect—a 200 nM BaP dose should be used as an initial treatment group in future experiments.
