Deposition and Patterning of Paraffin Layers for Culture-Based Detection of Mycobacteria in Environmental Samples

In this work, paraffin was deposited and patterned for culture-based detection of mycobacteria in environmental samples. To do so, a new method of depositing paraffin using butanol solvent to reduce viscosity of liquid paraffin during spin-coating deposition was explored. Using this approach, 18 mm glass wafers with paraffin layers of 10-70 µm have been successfully fabricated. In an alternative approach, paraffin was mixed with Opticlear solvent and 2-10 µm thick layers of paraffin were deposited on cycloolefin copolymer (COC) plastic wafers. Patterning of paraffin was accomplished in three ways. On glass, paraffin layers were patterned using either wet etching with Opticlear or dry etching using oxygen-based plasma. On COC, paraffin was patterned using lift-off with AZ4620 thick photoresist. The fabricated paraffin patterns were successfully challenged with mycobacteria to ensure that process paraffin layers were still viable. This work is a critical first step in the development of culture-based biochips that use paraffinophilic nature of microorganisms for rapid detection.e
