Abstract
Naive T cells differentiate into several different helper populations in response to cytokine milieu and pathogens. Regulation of this differentiation is important for mounting an effective immune response and preventing autoimmune diseases; for instance, abnormal Treg differentiation leads to tolerance against tumors and Th1 and Th2 differentiation may lead to autoimmune diseases and asthma, respectively. There are several studies showing that there is a correlation between epigenetic marks on signature genes and differentiation into distinct T cell lineages. Results from Wei et al. (2009) and Hawkins et al. (2013) show that positive H3K4me marks are enriched at the IL-4 locus in Th2 cells. Our preliminary data also show that the positive marks appear at lineage-specific cytokine genes in memory cells. Our aim is to determine whether this correlation reflects a causative relationship between presence of positive histone marks and T cells’ ability to express cytokines. In this study, we are planning to deposit histone modifications at the IL-4 gene in a locus specific manner. In order to do that, we are developing TALEMs, fusion proteins of TALE-based DBD and a catalytic domain of a histone modifying enzyme. Here we are describing development of TALE DBDs to target human and mouse IL4 genes and a set of experiments aiming at validating these TALE fusion proteins.
