Optimization of Methods for Generating Customized Gene-Edited Human Pluripotent Stem Cells

Human pluripotent stem cells (hPSCs) have the capacity for self-renewal and the ability to differentiate into any cell type providing an unlimited source of primary human cells capable of differentiation. Consequently, hPSCs have diverse applications ranging from novel lines for research in human development to the production of biological products for regenerative medicine. Improvements in gene editing technologies have expanded the potential applications of hPSCs, granting the ability to produce novel cell lines such as isogenic pairs of edited and unedited hPSC lines for gene function studies, cell lines with specific disease mutations, or reporter lines for functional studies and lineage tracing. Current methods for gene editing hPSCs are insufficiently robust for routine genome editing of diverse hPSCs. Costs and the time needed associated with optimization of standard gene editing practices may prove restrictive in many laboratories. The study reported in this thesis describe the optimization of standard CRISPR/cas9 genome engineering methods for of iPSCs, resulting in the development of a robust and broadly applicable procedure for routinely gene editing multiple iPSC lines.
