Building a better Placode: Modeling Neural Plate Border interactions with hPSCs

Ectodermal development has imperative steps with initial separation at early gastrula phases to a neural and non-neural zone with dorsally located Bone Morphogenic Protein (BMP) inhibitors(4). Between the neural and non-neural zone the neural crest and placode subtypes of the ectoderm form at intermediate BMP and WNT levels. However, whether the placode and neural crest arise from a common transient `border’ tissue or separately from the non-neural and neural zones respectively is still debated. While these events have been well tested using vertebrate model species, such as chick and zebrafish, a gap exists in our human specific understanding of these imperative early steps of ectoderm specification. Human pluripotent stem cells can be used to model the specification of the cranial placode and neural crest in vitro using inhibition of TGFB signals and low BMPs with WNT activation for neural crest and moderate BMPs for placode. This previous placode protocol is not sufficient to generate homogenous placode tissue and both models are limited in describing the complex three dimensional microenvironments in which these steps occur in vivo. This paper looks to achieve a more efficient monolayer differentiation of the placode lineage, and secondly develop a multicellular spheroid model of ectoderm develop to study the origin of the placode and neural crest tissues.
