Determining the Effect of HSP90 Inhibitor Geldanamycin on Herpes Simplex Virus Type-1 Production in Infected Vero Cells

Herpes simplex virus type-1 (HSV-1) is a highly contagious double stranded DNA virus that impacts at least one-third of the worlds’ population. HSV-1, or oral herpes, typically causes painful fever blisters, or cold sores, around the mouth and face. Invasion of HSV-1 into a host organism triggers a robust immune system response that eventually forces the virus into a latent period, which makes therapeutic options difficult to generate. Heat shock proteins (HSP’s) are universal proteins expressed during times of stress to cells, throughout which they facilitate protein assembly and regulate proteins within a host cell. Work with other viruses has shown that infection causes an increase in abundance of HSPs, which the viruses utilizes to ensure the proper assembly and conformation of viral proteins during infection. Previous results in this laboratory showed that heat shock protein 90 (HSP-90) increased in both transcription and translation during HSV infection. This project aims to determine the specific step of HSV replication which utilizes the HSP-90. In order to identify the critical time point during the HSV-1 replication cycle that HSP- 90 is required, an inhibitor of the HSP was added at hourly intervals following infection up to ten hours. The amount of virus produced was determined by plaque titration. The inhibitor used was Geldanamycin (GA) at 500 mM which inhibits HSP-90. The results of this study demonstrated that GA decreased virus production by 99.9 percent if added anytime time during the hours tested post-infection. These results suggest that HSP-90 plays a role throughout the HSV-1 replication and further work is required to demonstrate the efficacy of using HSP’s inhibitors in treating HSV infections.
