The following work describes new synthetic pathways toward one of the monosaccharide units, and analogs thereof, of the Staphylococcus aureus Type 5 and Type 8 capsular polysaccharides. Two main approaches to N-acetyl-D-mannosamine uronic acid were investigated. The first method utilized dithioacetal chemistry in which a pentose sugar was extended by one carbon atom via a nucleophilic addition to afford a hexose sugar derivative. The second synthetic procedure involved the addition of various nucleophiles to a selective protected D-mannosamine derivative to afford progenitors of D-ManAcA analogs.
