Abstract
It is known that unrecognized differences in tissue components among samples add noise to the measurement of gene expression when dissected cancer samples are used to correlate with clinical data in order to identify genes associated with disease status. The ability to accurately extract tissue components from clinical data has significant clinical importance in cancer diagnosis. The main goal of this project is to identify a statistical method with the high accuracy in estimating tissue components in tissue samples. CellPred and DeMix are two statistical methods widely used for determination of tissue contents. CellPred involves training on data with known pathological information using a multiple linear regression and estimating targeted tissue components, such as tumor, stroma, and other cell components in mixed samples. Demix estimates a targeted tissue proportion and specific gene expressions using a deconvolution algorithm. Each of these two methods has been utilized to extract the tissue components using both simulation data and clinic data. The fundamental principles of two methods are systemically studied and their performances in extracting tissue contents are investigated in the study.
