Molecular analysis of fs(1)polehole, a gene required for embryonic pattern formation and vitelline membrane integrity in Drosophila melanogaster

afs(1)ph is a gene whose function is required in two processes in oogenesis in Drosophila melanogaster. It is required for the production of an integral vitelline membrane which is necessary to prevent water loss and subsequent collapse of deposited eggs. It is also a member of the terminal class of genes which employ a signal transduction pathway to specify the terminal ends of the Drosophila embryo. This study has undertaken a molecular analysis of fs(1)ph to better understand how it carries out these functions. Sequences encoding fs(1)ph function have been identified and predicted to encode a protein glutamine-rich regions and homology to a domain found in the N-terminus of several proteins, including the Broad complex core proteins. This domain has no identified function but has been conserved in proteins of organisms ranging from viruses to mice. Unlike other proteins with this domain, the predicted fs(1)ph protein does not contain either zinc fingers or a carboxy terminal repeat. The fs(1)ph predicted protein also does not contain either a signal sequence or a transmembrane domain and is thus, likely to function intracellularly. This effectively eliminates models which postulate the peri-vitelline space as the site of action of the fs(1)ph protein. fs(1)ph encodes two transcripts found in ova ries and surprisingly, in testes and throughout embryogenesis. The unexpected presence of fs(1)ph transcripts in late embryos and testes may be indicative of additional functions of fs(:it 1)ph. To assess this, proven null alleles must be examined. A new fs(1)ph allele, generated in this study, is a small deletion of the fs(1)ph region, and may provide the means to determine the null phenotype of fs(1)ph.
