Development of Therapies for Treatment of Aggressive Cancers: Lessons from the Past and Current Perspectives

The development of effective chemotherapy regimens has had a tremendous positive impact on overall survival in patients with inoperable cancer. Despite these advances, developing new treatments to further improve survival and quality of life of patients remains a central goal of cancer research. Three independent projects highlighting different developmental approaches are presented here. First, a targeted therapy approach combining an antibody drug conjugate, ADCT-402, and the Bcl-2 inhibitor, Venetoclax, were tested in established leukemia and lymphoma lines. This combination proved to be synergistic through induction of apoptosis. Second, a series of compounds generated through a structurally based approach targeting ribonucleotide reductase (RNR) were shown to be effective against both a parental and gemcitabine resistant pancreatic cancer cell line. And third, a drug repositioning approach illustrated the potential of statins to reduce progression and metastasis in melanoma.
