Kinetic and Mechanistic Studies on the Reaction of the Reduced Vitamin B12 Complex Cob(II)alamin with Hydrogen Peroxide

Hydrogen peroxide (H2O2) is a strong oxidizing agent which is renowned for its damaging effects primarily due to the formation of the extremely reactive hydroxyl radical (*OH) upon its reaction with reduced metal complexes such as those of Fe(II). Kinetic and mechanistic studies of the reaction between the important reduced intracellular vitamin B12 complex cob(II)alamin (Cbl(II)) and H2O2 are reported. Rate constants, stoichiometric information on the reaction, and preliminary information on the intermediates and products of the reaction are presented. The second-order rate constant of the initial reaction between Cbl(II) and H2O2 was determined under three pH conditions and it is shown that Cbl(II) is rapidly oxidized by H2O2 in a pH independent reaction (pH 5-10). Through the use of *OH radical scavengers such as acrylonitrile combined with HPLC product analysis, our studies suggest that the initial reaction is a one electron oxidation of Cbl(II) by H2O2 to give aquacobalamin/hydroxycobalamin (cob(III)alamins, Cbl(III)) and *OH, followed by subsequent reactions of *OH with the corrin ring leading to the formation of complexes with spectra similar to the well-known stable yellow corrinoids. Possible reaction pathways are proposed.
