Lipid Raft TNF-a Pathway Analysis of Cytochrome C with Methylparaben and UV-B Treatment

Parabens are antimicrobials used in cosmetics as a preservative that prevents fungal and microbial growth. However, the side effects of parabens include: endocrine disruptive effects, increased activity of estrogenic receptors, sensitization with broken skin, and adverse reproductive effects. Methylparaben is the most commonly used paraben in topical skin products. UV-B radiation is known to increase the risk of skin cancer and is the most prevalent factor in skin cancer patients. In this study, M624 human melanoma cells were treated with methylparaben and UV-B light. It is shown that expression of the apoptotic TNF-a receptor is increased in the lipid rafts of the M624 cells treated with 5mM of methylparaben, but not in M624 cells treated with 5mM methylparaben and 25mJ/cm2 UV-B light. The purpose of this research was to study the expression of the cytochrome c protein in relation to methylparaben and UV-B treatment. It is shown that cytochrome c expression is decreased in the 5mM methylparaben M624 cell sample in contrast with an increased expression of TNF-a in the 5mM methylparaben sample. Caspase-3 expression was also observed to decrease in the 5mM methylparaben treated sample. Results indicate that 5mM methylparaben and UV-B treatment cause apoptosis through a lesser known extrinsic apoptotic pathway that does not involve cytochrome c release or caspase-3 activation.
