Modular Clickable and Photoactive NAADP Analog for Isolation and Purification of the NAADP Receptor

Nicotinic acid adenine dinucleotide phosphate (NAADP), a derivate of nicotinamide adenine dinucleotide phosphate (NADP), has been shown to be a potent intracellular second messenger that triggers calcium release from intracellular stores at nanomolar concentration in a variety of biological systems. However, the NAADP binding protein has not yet been identified unambiguously and the regulation of NAADP mediated Ca2+ release is still controversial. To address this issue, we synthesized a bifunctional “all-in-one-click” NAADP analog in which 3-azido-5-ethynylbenzoic acid was attached to the amino group of 5-(3-aminopropyl) NAADP to form an NAADP analog which was a clickable photoaffinity label. This NAADP analog was synthesized by coupling 3-azido-5-ethynylbenzoic acid to ethyl 5-(3-aminopropyl)nicotinate using a PyBOP coupling agent. The nicotinic acid ester was then hydrolyzed, and the nicotinic acid derivative was incorporated into NAADP using the pyridine base exchange reaction catalyzed by E98G Aplysia californica ADP-ribosyl cyclase. The synthesized NAADP analog was shown to begin to induce Ca2+ release from sea urchin egg homogenates in the millimolar concentration range and to induce sub-threshold desensitization of Ca2+ release with an IC50 of approximately 0.3 mM.
