Abstract
In the US alone an estimated 10-15% of couples are considered infertile as defined by World Health Organization (WHO) criteria. Among them, it is estimated that 30% of these couples are infertile due to the male partner (The National Women’s Health Information Center, NWHIC). Male infertility can be due to a number of deficiencies including immotile sperm. However, the basic cellular and/or physiological mechanism(s) that regulate sperm motility, especially those involving membrane transporters, are not well defined. Recent work on the Na,K-ATPase alpha4 isoform has shown that this transporter plays an important role in maintaining rat sperm motility possibly because it is required to regulate the intracellular pH of sperm due to its effects on the Na/H exchanger (NHE). More than one isoform of both the Na,K-ATPase and the NHE are found in sperm leaving open the question as to the individual roles that all of these isoforms have in sperm motility. Therefore, this dissertation will focus on determining the functions of the individual Na,K-ATPase alpha isoforms in sperm motility regulation. In addition this work examines the possibility that the Na,K-ATPase alpha4 isoform is important for clearing protons leaking from metabolically active mitochondria out of the sperm. Finally, the expression of the Na,K-ATPase alpha4 isoform protein in humans is also explored as it is likely that this protein has a significant role in human reproductive biology.
