Abstract
Quantitative analysis in infrared microscopy was studied. Various physical phenomena, caused by sample structure, refractive index differences, and angles of incidence were investigated. Such phenomena include diffraction and refraction, which introduce spectral artifacts that can limit quantitative infrared microspectroscopy. The address the limiting phenomena, improvements in microscope design and the optimum use of sampling accessories were sought. Thus, a newly developed imaging detector was used along with an attenuated total reflectance (ATR accessory, providing sub-wavelength spatial resolution, on the order of 4 micrometers. Moreover, smaller samples were interrogated without obvious spectral artifacts due to sample structure. In addition, a specially-designed macro ATR accessory was created for the collection of spectral images from surfaces of hard-to-prepare samples, which included living tissue and a biological sample. Finally, the use of ATR and Raman spectroscopy was successfully applied, laying the groundwork for development of a combined Raman/Infrared ATR microscope.
