SYN3 in Chloroplasts of Arabidopsis thaliana: Effects of Knockdown and Overexpression and Localization Techniques

Cohesins are complexes of proteins involved in sister chromatid cohesion during mitosis and meiosis as well as DNA repair and transcriptional regulation. One subunit of this protein complex, called SYN3, has been found in chloroplasts of Arabidopsis thaliana. In an effort to determine the role of SYN3 in chloroplasts, we examined RNAi knockdown and overexpression lines. Our results indicate that knockdown and overexpression of SYN3 do not affect chloroplast size or number, but knockdown does affect senescence, with earlier senescence, disruption of chloroplast structure, and fewer autophagosomes. Localization shows SYN3 distributed throughout the chloroplast stroma, but no co-localization with DNA. Several methods were used to localize SYN3 for TEM. High paraformaldehyde and low glutaraldehyde concentrations in sodium phosphate buffer and embedment in LR White preserved ultrastructural integrity and retained antigenicity. Post-section staining with osmium tetroxide vapors allowed for visualization of the thylakoids within the chloroplast without introducing artifacts.
