Enhancer identification and activity evaluation in the red flour beetle, Tribolium castaneum

Changes in the gene regulatory networks (GRNs) are the major driving force of morphological diversity. Evolution of cis-properties (non-coding DNA elements such as enhancers) often plays important roles in the production of diverse morphology, as changes in cis-properties tend to be less deleterious. However, studies on enhancers in species outside of established model systems are often limited because of the lacking a well-established enhancer identification and evaluation strategy. In this study, I used FAIRE (Formaldehyde Assisted Identification of Regulatory Elements) to identify enhancers in the red flour beetle, Tribolium castaneum. Furthermore, by using a construct with the Drosophila synthetic core promoter (DSCP), I established a functional reporter assay system in Tribolium. This reporter assay system, in combination with FAIRE-based chromatin profiling, will make the research on enhancers more accessible, which will provide us with more insights into the evolution of the regulatory mechanisms underlying morphological diversity.
